What thickness of acrilic would I need to make a tank thats 45x12x24,
holding at least 500 lbs.

It kind of depends.
The number varies dramatically, depending on if the acrylic is structural,
or other stuff (steel/...) is being used to support the acrylic.
Braces across the top can also dramatically change the numbers.
And as acrylic is not cheap, do you want all sides acrylic, or only one?

all sides, or at least 2 front/back. Ok what about plexi glass thickness?

Plexiglass is a brand of acrylic. We would still need to know though
what you want for top bracing. If you have a full top with small
cutouts, 3/8's would be fine, if you wanted a fully open top with no
noticable bending you would need 1 inch or thicker.

fully open with 2 braces across, also it would be in a wooden frame.
